This BIORESEARCH TECHNOLOGY UPDATE is to inform you about the use of the Warner Instruments TC-344B Dual Channel Temperature Controller to determine the relationship between adenomatous polyposis coli (APC) protein with microtubule (MT) ends and its binding partner EB1.

The TC-344B Temperature Controller was used to maintain the temperature of the Warner SH-27B Inline Solution Heater and Warner RC-30 Imaging Chamber during drug perfusion of living canine kidney epithelial cells and mouse primary fibroblasts.

Using the APC-deficient cell line and small inhibitory RNA inhibition of EB1 expression, the study demonstrated that the association of endogenous APC with MT ends correlates directly with their increased growth stability, that this can occur independently of its association with EB1, and that APC and EB1 can associate with MT plus ends by distinct mechanisms.

The dynamic MT cytoskeleton is critical to cell functions such as cell division, directed cell migration, the immune response, and neuronal development.
